Metalloproteinase Inhibition and Erythroid Potentiation Are Independent Activities of Tissue Inhibitor of Metalloproteinases - l

چکیده
Tissue inhibitor of metalloproteinases-l (TIMP-l), the major physiological matrix metalloproteinase inhibitor and a potent antimetastatic factor, also stimulates the growth of erythroid progenitors (erythroid-potentiating activity). We analyzed the relationship between the growth factor activity and protease inhibition by preparing purified TIMP-1 “knockout” proteins lacking in vitro antiproteolytic activity. The growth-stimulatory effect of these N-terminal TIMP-1 point mutants, as tested in an in vitro assay using erythroid precursors (erythroid burst-forming units) was equal to that of unmutated TIMP-1. A fully antiproteolytic C-terminal TIMP-

Introduction: Metalloproteinases and their tissue inhibitors play an important role in the metastases formation. A multistage process of carcinogenesis requires the involvement of numerous enzymes and compounds that facilitate the expansion of tumor cells. The formation of metastases depends on both metalloproteinases and tissue inhibitors activation leading to the acti...

Matrix metalloproteinases (MMP) are crucial for homeostasis (tissue remodelling and repair, bone growth, wound healing, etc.) and pathology (metastasis, angiogenesis, aneurysm rupture, etc.). Upregulated MMPs from macrophages are thus a two-edged sword, playing both defensive and aggressive roles. The related family of ADAMs (a disintegrin and a metalloproteinase) is sometimes overlooked becaus...

Development of the embryonic kidney results from reciprocal signaling between the ureteric bud and the metanephric mesenchyme. To identify the signaling molecules, we developed an assay in which metanephric mesenchymes are rescued from apoptosis by factors secreted from ureteric bud cells (UB cells). Purification and sequencing of one such factor identified the tissue inhibitor of metalloprotei...

Matrix metalloproteinases (MMPs) play a central role in many biological processes such as development, morphogenesis and wound healing, but their unbalanced activities are implicated in numerous disease processes such as arthritis, cancer metastasis, atherosclerosis, nephritis and fibrosis. One of the key mechanisms to control MMP activities is inhibition by endogenous inhibitors called tissue ...
